Comparisons of a Formaline Substitution with Routine Chemical Fixatives on Cryosections

چکیده

This study was designed to compare tissue preservation efficiency of formaldehyde substitution (FS) on cryosections with acetone (AC) and formaldehyde (FA). Brain, kidney, heart and liver had obtained from adult Dutch rabbit and were fixed with either FA or FS and AC prior to cryosectioning. Hematoxylin and eosin staining showed that AC is a poor fixative in preserving the general tissue and cellular organization. FS and to a lesser extent FA, produced significantly better results. All of cryosections were stained by hematoxylin and eosin and were studied by light microscopy. Results demonstrated that, overall, FS is the best fixative tested. However, FA fixation gave poor results in preserving neuronal tissues. Overall, the data suggest that AC fixation is unacceptable for preservation of most samples, whereas FA and FS fixation should be chosen according to the tissues and proteins to be studied.

Post-embedding immunoelectron microscopy of tissue processed by rapid freezing and freeze-substitution without chemical fixatives.

With great interest we read the recent article by Usuda et al. (1), which describes the immunoelectron microscopy of tissue processed by rapid freezing and freeze-substitution without chemical fixatives. In their article, Usuda et al. say that “our result . . . should (sic) be the first demonstration of immunostaining of rapidly frozen tissue processed without chemical fixation.” We would like ...

Immunoreactivity of glutamate in mouse retina inner segment of photoreceptors with in vivo cryotechnique.

The purpose of this study was to clarify a previously controversial issue concerning glutamate (Glu) immunoreactivity (IR) in the inner segment (IS) of photoreceptors by using in vivo cryotechnique (IVCT) followed by freeze substitution (FS), which enabled us to analyze the cells and tissues reflecting living states. Eyeballs from anesthetized mice were directly frozen using IVCT. The frozen ti...

Evaluation of two commercial and three home-made fixatives for the substitution of formalin: a formaldehyde–free laboratory is possible

BACKGROUND Formaldehyde (HCHO) is a gas (available as a 37% concentrated solution, stabilized with methanol). The 10% dilution (approximately 4% formaldehyde) has been used as a fixative since the end of the 19th century. Alternative fixatives are also commercially available or may be prepared in-house in laboratories. Statements by the IARC, along with other USA agencies (CalEPA, RoC/NTP) on t...

Tandem high-pressure freezing and quick freeze substitution of plant tissues for transmission electron microscopy.

Since the 1940s transmission electron microscopy (TEM) has been providing biologists with ultra-high resolution images of biological materials. Yet, because of laborious and time-consuming protocols that also demand experience in preparation of artifact-free samples, TEM is not considered a user-friendly technique. Traditional sample preparation for TEM used chemical fixatives to preserve cellu...

LYMPH NODE REVEALING SOLUTION: A NEW USEFUL METHOD FOR ISOLATION OF MINUTE AXILLARY LYMPH NODES IN BREAST CANCER

Breast cancer is one of the most common malignancies in the world. The most important prognostic factor in this cancer is the stage of the tumor. Detection of lymph nodes with or without malignancy is essential for determining the stage of the tumor and setting a therapeutic plan. There are different methods for isolating lymph nodes in surgical specimens of breast cancer in the pathology ...
